2. Entecavir is a selective and effective HBV inhibitor. EC50 value in HepG2 cells is 3.75 nM .
3. Entecavir is a guanine nucleoside analog and has inhibitory effect on hepatitis B virus (HBV) polymerase.
4. Entecavir can inhibit all three activities of viral polymerase (reverse transcriptase): (1) the start of HBV polymerase; (2) Formation of reverse transcriptional negative strand of pregenomic mRNA; (3) Synthesis of HBV DNA positive strand. The inhibition constant (Ki) of entecavir triphosphate on HBV DNA polymerase is 0.0012 μ M. Effects of entecavir triphosphate on cellular α,β,δ DNA Polymerase and Mitochondria γ The inhibition of DNA polymerase is weak, Ki value is 18 to 160 μ M.
